I am investing yearly Rs. 20,000 in Reliance ULIP, Rs. 2,000 in monthly SIP in Direct Growth Option Plan of ICICI Prudential Focused bluechip equity and Rs. 2,000 in monthly SIP Direct Growth Option Plan of Mirae Asset Emerging Bluechip Regular. Should I change my third investment with HDFC midcap options?

You don’t have to change your 3rd option, continue with it. If you are really interested in improving your portfolio then I will suggest you to change your 1st option with midcap opportunities capable of becoming large cap in future. Reason being ULIPS have a low rate of return.
